Scotland have lost just one of their last 15 matches against Italy, but that came on their final visit to the Olympic Stadium in 2024.
It was another Scottish collapse in the second half, as the home side came from behind to win 31-29.
Townsend is not underestimating the Italians as the Scots aim to start well before facing England at Murrayfield and Wales in Cardiff.
“It is difficult to play away from home against a more competitive team,” Townsend said.
“We’ve had some tough battles with them. They beat us in Rome two years ago. They’re selling out in this stadium now. It’s a really loud atmosphere.
“They have a cohesive squad, players who have played together for a number of years, and really talented individuals. We know it’s a very tough game.”
Before speaking about his latest century, Townsend says he never expected to remain in the role for so long, while insisting his thirst for the role remains.
He also played down a report suggesting he had agreed to take over as Newcastle Red Bulls coach after the 2027 World Cup.
The 52-year-old added: “I remember when I first started coaching and especially when I became head coach at Glasgow, I said to myself: ‘I love this. “I will give everything, and if it doesn’t work out, it wasn’t meant to happen.”
“I’ve tried to have that mentality all the time. It’s something I love doing, but I feel very proud and lucky to have had the opportunity after my playing career.”
