Australia resumed the match on 96-3, trailing by 102 runs, with Sutherland and Perry putting the home side on 133 in a strong position before the latter was dismissed by Deepti Sharma.
Wicketkeeper Beth Mooney took out 19 off 53 balls to help Australia go in front and provide support to Sutherland, who played admirably on an increasingly challenging surface.
She was unbeaten on 93 at tea and quickly moved on to her historic century before bowling off Deepti.
Alana King and Lucy Hamilton put together 34 to give Australia a huge lead and then a tired India faded under the lights in the final session.
The visitors fell to 10-2 and then Sutherland claimed the key wickets of Jemima Rodrigues and skipper Harmanpreet Kaur to leave India 64-4.
Left-arm fast bowler Hamilton, making her Test debut, removed Deepti and Richa Ghosh in the space of three balls to leave India reeling at 82-6 and in danger of losing the match within two days.
However, Pratika Rawal’s challenging 43 not out and Sneh Rana’s unbeaten 14 ensured that the Test would move to the third day.
