De Minaur, who was hoping to reach his first Grand Slam semi-final for the seventh time, had not beaten Alcaraz in five previous attempts.
In a close opening set, De Minaur stunned the crowd with powerful strikes as he tried to match the speed and intensity of the world number one.
He fought his way back from two breaks before the top seed hit the winner at 6-5 after about an hour of play.
Pat Cash, a former Wimbledon winner, said the first set showed “some of the highest levels of tennis I’ve ever seen.”
“There were a couple of players who were refusing to back down and were hitting balls from two or three feet behind the baseline,” Cash told BBC Radio 5 Sports Extra.
“It was like a video game, going back and forth. It was very fast; there were lob shots, high defensive balls, chasing backs, there was everything. An unbelievable combination of tennis.”
But the gap in quality quickly became apparent as Alcaraz raised his game and De Minaur struggled to serve the six-time Grand Slam winner’s serve.
“Once we started asking could he [De Minaur] “He kept the bar high, the wheels fell off and Alcaraz didn’t back down,” Cash added.
