National League South side Slough Town were the first team drawn and will host Macclesfield of the National League North.
Chelmsford City, who dispatched local rivals Braintree Town 4-1 at the weekend to reach the second round, have a home draw against fellow National League South club Weston-super-Mare.
Elsewhere, Bristol Rovers boss Daryl Clarke returns to his former club Port Vale and National League side Gateshead host League Two leaders Walsall.
Following a dramatic 4-3 win over Forest Green Rovers on Friday, Jack Wilshere’s Luton Town travel to the North West Coast for a draw at Fleetwood Town.
Brackley Town play at home to League One side Burton Albion, while Wealdstone visit National League side Grimsby Town.
2013 winners Wigan Athletic, who needed a penalty shootout to beat Hemel Hempstead Town in the first round, face a home draw against League Two Barrow.
After their extra-time heroics, Buxton FC moved from the National League North to struggling Cheltenham Town in League Two.
The confrontations will take place over the weekend from December 6 to 7.
Crystal Palace, the defending champion, and the rest of the clubs playing in the Premier League and Championship will enter the competition in the third round in the new year.
